Purify up to 25 µg of high-quality plasmid DNA from 1-5 mL bacterial cultures in less than 30 minutes using the alkaline-lysis method and HiBind® Mini Column technology.

pGEM plasmid was purified from 4 mL DH5α cultures harboring the plasmid and eluted in 50 µL volume using kits from Omega Bio-tek and Company Q according to manufacturer’s recommended protocols. Plasmid DNA concentration was determined by optical density measurements using Thermo Scientific’s NanoDrop™ 2000c system.

E.Z.N.A.® FastFilter Plasmid DNA Mini Kit

Purify high-quality plasmid DNA from up to 5 mL of culture following the alkaline-lysis method using a first-of-its-kind FastFilter Mini Column nested within a regular HiBind® DNA Mini Column to combine lysate clearance and DNA binding into one or two centrifugation steps depending on the culture volume.

E.Z.N.A.® Endo-free Plasmid DNA Mini Kit II

Purify up to 70 µg endotoxin-free plasmid DNA from 5-15 mL bacterial culture in less than 30 minutes using integrates an efficient endotoxin removal step into the plasmid purification procedure to produce high-quality transfection grade (<1 EU/µg) plasmid for efficient transfection.

Quality and yield of plasmid DNA purified using the E.Z.N.A.® Endo-Free Plasmid DNA Mini Kit. Plasmid DNA was isolated from 3 mL of three different bacterial cultures. DNA was eluted in 100 µL endotoxin-free elution buffer and quantitated using Thermo Fisher NanoDrop® 2000 and dynamic turbidity method.

Mag-Bind® Endo-free Plasmid Mini Kit

Purify endotoxin free (<0.1 EU/µg) plasmid DNA using magnetic beads from up to 1.5 mL culture volume.

pGEM plasmid DNA was purified from 1.5 mL DH5α cultures (n=3) using the Mag-Bind® Endo-free Plasmid Mini Kit, as well as Company P’s Kit, following manufacturers’ recommended protocol. Purified Plasmid DNA was quantified using Thermo Scientific’s NanoDrop™ 2000c system. Plasmid preparation was not only endotoxin-free (<0.1 EU/μg) for Omega Bio-tek’s Kit, but the yield was also significantly higher compared to Company P’s Kit.

Isolate up to 200 µg of high-quality plasmid DNA from 15-50 mL bacterial cultures in less than 40 minutes using Lysate Clearance Filter Syringes instead of centrifugation to rapidly clear bacterial lysates post-alkaline lysis.

Plasmid DNA was isolated from eight 30 mL bacterial cultures grown 16 hours in LB medium using the E.Z.N.A.® FastFilter Plasmid DNA Midi Kit and eluted in 300 µL elution buffer. Plasmid DNA (1% of total purified DNA) was analyzed on a 0.8% agarose gel. Lanes 1-8 represent plasmid DNA from 8 different bacterial cultures and Lane M represents the 1 kb DNA ladder.

E.Z.N.A.® Endo-Free Plasmid DNA Midi Kit

Extract plasmid DNA from 15-50 mL of bacterial cultures in LB medium to produce up to 200 µg of high copy number plasmid DNA and process up to 100 mL of culture when working with low copy number plasmid.

pEGFP-N1 plasmid was purified from 50 mL DH5α cultures harboring the plasmid using Omega Bio-tek’s E.Z.N.A.® Endo-Free Plasmid DNA Midi Kit following manufacturer’s recommended protocol. Plasmid DNA was eluted in 500 µL volume and quantified using Thermo Scientific’s NanoDrop™ 2000c system. The Endotoxin concentration was estimated based on a Limulus Amoebocyte Lysate (LAL) assay.
